Platypus Technologies has developed a product line that gives you more control over designing a cell migration assay.

Each Oris Cell Migration Assembly kit contains the Oris Cell Seeding Stoppers for creating a detection zone at the center of each well in a 96-well plate. Since the stoppers are not pre-inserted into the wells, researchers have two options; coat the plate with an extracellular matrix component or use the coated plate provided to design an assay. Create unique 3-dimensional overlays in each well and watch how cells invade and respond to various compounds (chemokinesis). The Oris Universal Cell Migration assembly Kit is an ideal alternative to the scratch assay for studying wound healing. The detection zone created by the Oris stopper provides a consistent area for a 2-D closure assay.

The original Oris Universal Cell Migration Assembly kit is supplied with a 96-well, black, clear-bottom plate, an Oris Detection Mask, an Oris Stopper Tool, and 96 Oris Cell Seeding Stoppers.*

The NEW Oris Cell Migration Assembly Kit-FLEX now allows for more control over how many tests can be performed in a single experiment. Now use anywhere from 4 to 96 test wells per experiment; it is your choice. Whether you are studying cancer metastasis or wound healing, the Oris Cell Migration Assembly Kit-FLEX adapts to your unique experimental design.

1. Insert Oris™ Cell Seeding Stoppers

2. Seed each well with cells (to achieve 90-95% confluency around Oris™ Stoppers)

3. Incubate plate to allow cell adherance to bottom of well

4. Remove Oris™ Stoppers

5. Allow cells to migrate into the detection zone

6. Stain cells

7. Measure migration with a microplate reader, microscope or digital imaging system

1. Coat Oris™-compatible, 96-well plate with choice of ECM (unless already coated)

2. Insert Oris™ Cell Seeding Stoppers

3. Seed each well with cells (to achieve 90-95% confluency around Oris™ Stoppers)

4. Incubate plate to allow cell adherance to bottom of well

5. Remove Oris™ Stoppers

6. Overlay adherent cells with ECM

7. Allow cells to invade into the detection zone

8. Stain cells

9. Measure invasion with a microplate reader, microscope or digital imaging system

 

Manually inserting Oris Cell Seeding Stoppers will result in data with higher CV values than data obtained using the Oris Cell Migration Assay kits pre-populated with mechanically inserted stoppers.
