For in vitro experiments aggregation is one of the major issue affecting protein functions, such as : folding, ligand binding, enzymatic activity, interactions….

Despite being frequently used for determining protein purity, SDS-Page cannot tell you the whole story, and don’t appear as the best option to determine the aggregation state of your protein model.

The data below demonstrates the importance of evaluating aggregation of your protein of interest. While both of these PD-1 proteins are over 90% pure by SDS-PAGE and Coomassie staining, they exhibit very different profiles by FPLC. The protein on the left shows significant aggregation, while the protein on the right is less than 5 % aggregated.
