Secrete-Pair™ Gaussia Luciferase Dual Luminescence Assay Kits

The Secrete-Pair™ Dual Luminescence Assay Kit is designed to analyze the activities of Gaussia Luciferase (GLuc) and Secreted Alkaline Phosphatase (SEAP) in promoter reporters (Gluc-ON reporters)  and miRNA target validation reporters (miTarget reporters).

Both GLuc and SEAP are secreted proteins, allowing detection without cell lysis, just from the cell culture supernatant. GLuc reveals the specific activity of the cell-based assay and the SEAP allows transfection normalization, avoiding the need for cell line establishment to get reliable results.

Transient transfection of the Gluc/SEAP reporter with Endofectine Max quickly provides a convenient and reliable cell-based screening assay.

Simple way to get high quality data

With a direct analysis of the supernant, without cell lysis, the same sample can be assayed multiple times for different time points. It helps a lot to define the best time point for screening ensuring the best results. It also reduces sample-to-sample variation

Fast development of cell-based assays

With a dual reporter, data are normalized from point to point. This makes transient transfection a reliable and fast method to set a cell-based screen assay.

Flexible acquisition of higher range

GLuc is 1,000 times more sensitive than firefly and Renilla luciferases bringing higher range of values. Taken together with the 2 robust buffer conditions (provided in the kit), it is possible to explore very low GLuc expression as well as very high expression.
