Gold standard one component TMB

18/01/2019 by Dimitri Szymczak, PhD No Comments

Tetramethylbenzidine (TMB) is a classic substrate for peroxidase activity that is used for microwell plate analysis such as an ELISA test. A quantitative color change from yellow to blue reveals the activity and can be read at 620-650 nm for ELISA test.

Substrate selection in immunoassay development

Surmodics have developed several types of TMB, for different purposes.

How to produce high yield capped mRNA

17/01/2019 by Dimitri Szymczak, PhD 2 Comments

Messenger RNA can be used for protein expression into the cells. The delivery of mRNA is the delivery of a new function that can activate a biological process. In the particular case of the CAS9 mRNA, it is simple way to make gene editing. Messenger RNAs are at the center of attention to fight cancers and for personalized medicine. However, production of capped mRNA can be a challenge. Let’s see how to face up to it here with some tips.

Achieve success with RNA-based CRISPR-CAS9 knock-out

15/01/2019 by Dimitri Szymczak, PhD No Comments

RNA-based CRISPR-CAS9 gene editing is a vector-free approach that is required for therapeutic perspectives. However, from a practical point of view, what appears as a major challenge to engineer primary cells, is actually also an efficient and smart solution for numerous other knock-out projects on cell lines. We see here why and how.

Luciferase promoter reporter clones

08/01/2019 by Dimitri Szymczak, PhD No Comments

Cell-based assays, screening for pathway activation or inhibition are classically done with promoter reporter clones expressing Firefly luciferase. Brighter, more stable, more sensitive, more convenient and cheaper, Gaussian luciferase has become popular over the past years. Let’s discover here the next generation of reporter clones, called Gluc-ON.
