Analyte quantification (in pg/ml or ng/ml) such as IL-6, TNFalpha and numerous other proteins is conveniently performed with immunoassays called ELISA kits. “Best”, in this context, means a smooth, rigorous and reliable means in order to get data in which we can trust, with high reproducibility.
Just one file for fast and accurate protein quantification
The method exists. It is based on 4PL and 5PL regression, and it requires quite a lot of gymnastics with maths. So, I have built an analysis tool that includes all the formulas and the method.
You just need to enter into the file the raw data (OD) and the 4PL and 5PL parameters (provided by scanner software or GraphPad for example), and then you get final concentrations of your favorite analytes.
By the way, thanks to the standard duplicates, we can choose between 4PL and 5PL based on a simple %fitback calculation, also included in the tool. It’s kind of quality control for each point and it helps define the most appropriate regression between 5PL and 4PL case by case.
