ADME-Tox: liver fractions with High Aldehyde Oxidase and Xanthine Oxidase Activity

Sekisui XenoTech has just established a method for high Aldehyde Oxidase and Xanthine Oxidase activity in pooled Human liver test systems.

These new test systems (Pooled Human Liver S9 and cytosol) have been prepared from tissue treated to preserve high Aldehyde Oxidase (AO) and Xanthine Oxidase (XO) activity, yet without significantly impacting major cytochrome P450 (CYP) activity.

New range of Pig P450s Bactosomes

If you are looking for enzymatic in vitro tools for drug metabolism, reaction phenotyping, metabolite generation, using pig as model, you may find these newly released Pig Bactosomes from Cypex of interest.

Bactosomes are recombinant enzymes expressed in E. coli with a patented expression system that enables expression of drug metabolising enzymes without the need for large modifications to be made to the proteins. They demonstrate excellent batch-to-batch consistency and robust activity levels. Each batch is characterized for linearity with time and CYP concentration, Km and Vmax.

New Ready-to-use Dermal Subcellular Fractions

Human and Animal Dermal S9 Fractions now available as standard products

Subcellular fractions are widely used in drug discovery and preclinical drug development to evaluate the in vitro metabolism of new therapeutics. Skin fractions are already available from tebu-bio as catalogue items for CD1 Mouse and IGS Sprague Dawley Rat.

In collaboration with Sekisui XenoTech, tebu-bio is now adding human and minipig dermal subcellular fractions to the list of species available as standard test systems.

Toxicity Assessment of Co-Culture Human Systems

The inadequacy of animal models to predict human biology in the drug development process is becoming increasingly clear, due to species differences in uptake and metabolism at both cellular and organ levels.

As a result, there is a need for more human model systems to be incorporated earlier in research and development.

Innovative concepts such as “body on a chip” have been introduced, but the complexity and miniaturization of many of the formats has limited applicability on a commercial scale.

SciKon is developing tools that better recapitulate biological systems in bench-top cell culture formats, which are amenable to mass manufacturing (introduced recently in the post Cell Signaling isn’t static…your cell culture shouldn’t be either!).
