RNA Interference (RNAi) is one of the most exciting discoveries of the past decade in functional genomics and proteomics. Silencing becomes a powerful tool for studying the gene function.
Ready-to-transfect siRNA for a transient silencing
The first generation of silencing tools was the chemical siRNA allowing only a transient silencing.
tebu-bio are currently offering over 5,000 target-specific 20-25 nucleotide long siRNAs that can be used to knockdown protein expression in a broad variety of mammalian cell types (Santa Cruz Biotechnology).
Chimera RNA interference (chimera RNAi) is a process by which small interfering RNA/DNA chimera triggers the destruction of mRNA. We have over 14,000 pre-design Chimera RNAi in catalogues (Abnova).
Pre-designed shRNA expressing vectors
Later, the second generation appeared in the market was shRNA expressing vectors, allowing the extinction of gene expression transiently or stably. The SureSilencing™ pre-designed shRNA plasmids specifically knock down the expression of every human, mouse or rat gene by RNAi (SA Biosciences). For each gene, we provide four plasmids each with a different pre-designed short hairpin RNA (shRNA) sequence. Unlike chemically synthesized siRNA, plasmid-based shRNA provides a renewable source of RNA interference. You can amplify enough plasmid in your own lab to complete your project.
Target-specific silenced and ready-to-use cell lines
The third generation of silencing tools is the access to cell lines already silenced for your gene of interest. This tool is only available at tebu-bio: SilenciX.
SilenciX cell lines provides a model to quickly check if the silencing of the suspected target is good for the work hypothesis or not.
Knock-down effects assessment
To assess the knowdown effect of your target-specific siRNA, an exhaustive choice of tools is at your disposal: pathway specific macroarrays to check which genes are activated or inactivated in a particular cellular pathway, PCR primer sets to validate the target specific mRNA degradation, antibodies to assess protein expression and much more...
