Rat Osteoblasts (ROb)

 

Rat Osteoblasts (ROb) are isolated from rat bone and are cryopreserved at second passage. ROb can be passaged at least 10 population doublings. ROb are a powerful tissue engineering tool for the study of signaling pathways and 3-D modeling. Sustained delivery of basic fibroblast growth factor (bFGF) enhances bone formation following osteoblast transplantation1. Prostaglandin E increases fibronectin and promotes bone formation in ROb via the EP(1) receptor, phospholipase C, PKCalpha, and c-SRC signaling pathway2. Via the FGF2 pathway, prostaglandin F2-alpha regulates ROb metabolism3. Osteoblasts express A disintegrin and metalloprotease-1 (ADAMTS-1), which breaks down extracellular matrix molecules and is expressed at regions of new bone formation4. ADAMTS-1 is a target for parathyroid hormone in bone5. Hypoxia decreases the growth, differentiation and bone-forming capacity of ROb6. ROb can be used in tissue engineering models. Rat osteoblasts have been cultured in 3-D matrices, including processed bovine cancellous bone which resulted in osteoblast differentiation7. Plasma-treated porous polylactide membranes increases the attachment and growth of ROb for the use of tissue engineering8.  

Each lot tested negative for HIV, Hepatitis B and Hepatitis C and negative for mycoplasma, bacteria, yeast, fungi.
